Description:
DNAHomopolymericTailingKitprovidesqualifiedreagentsfortheadditionofhomopolymertailstothe3'endsofDNAwithterminaldeoxynucleotidyltransferase(TdT).Underoptimizedassaycondition,approximatelyaverageof30~70ntoligo(dA)andoligo(dT)or15~45ntoligo(dC)andoligo(dG)couldbeaddedtothetargetsubstrate.
TdTisatemplate-independentDNApolymerasethatcatalyzestherepetitiveadditionofdeoxynucleotidestothe3'hydroxylterminusofDNAmolecules.TheenzymewasgeneratedfromanE.colistrainthatcarriestheclonedTdTgenefromcalfthymuswithselectedmutations.Protruding,recessedorblunt-endeddoubleorsingle-strandedDNAmoleculesserveasasubstrateforTdT.TheadditionofdNTPsto3′-overhangingendsismoreefficientthanwith3'-recessedorbluntends.TdTincorporatesdATPanddTTPwithhigherefficiencythandCTPanddGTP.Theoptimizedmastermixturestimulatesthetailingofthe3'-endsofDNAfragments,evenapplicableforincorporatingribonucleotidesandmodifiednucleotides(e.g.,fluorescein-,biotin-,aminoallyl-labelednucleotidesanddideoxynucleotides).
